环状芽孢杆菌A1.383产酵母胞壁溶解酶发酵条件的研究

摘  要: 环状芽孢杆菌A 1 3 83产细胞壁溶解酶的适宜的碳源为酵母葡聚糖 ,氮源采用蛋白胨与(NH4 ) 2 SO4 混合氮源 ,接种量 6%~ 1 0 % ,2 5 0mL三角瓶装液量为 40mL ,发酵温度为 3 0~ 3 5℃ ,培养基初始pH 6 5~ 7 0。在此条件下发酵 5 4h后可达到最高酶活 74 6u/mL ,比条件优化前的酶活提高了 3 8 7%。将所得的发酵液用于红法夫酵母虾青素的酶法提取 ,总类胡萝卜素的提取率达到 96%以上。 The effects of different factors on lytic enzyme production by Bacillus circulans A 1.383 were studied in flask. It is found that water insoluble yeast glucan was the best C sources and the mixture of peptone and (NH_4)_2SO_4 was the best N sources. It is also seen that the best inoculum concentration was 6%~10% with 40?mL medium volume in 250?mL flask for B. circulans A 1.383 to produce lytic enzyme with high activity. The fermentation temperature was 30~35℃ and the initial pH value of growth medium was 6.5~7.0 in this paper. The lytic enzyme activity of fermentation fluid reached 74.6?u/mL and it is increased by 38.7% compare with the initial. The fermentation fluid was used for extracting astaxanthin of Phaffia rhodozyma and over 96% of the total carotenoid could be extracted within 36 h when reaction temperature was 30℃ and pH was 6.5.
