摘 要: 利用基因工程技术生产1,3-丙二醇,特别是途径工程的利用已取得了重大突破.利用生物技术生产1,3-丙二醇的成本比化学法的低25%,它将逐步实现工业化生产.介绍了一步法生产1,3-丙二醇的超级基因工程菌所需的3-磷酸甘油脱氢酶(GPD1)、甘油3-磷酸酶(GPP2)、甘油脱水酶(dhaB)和1,3-丙二醇氧化还原酶(dhaT),并对关键酶dhaB进行了分析,同时介绍了穿梭质粒和甘油-3-磷酸酶积累对生产的影响.最后,对今后的研究重点和策略进行了探讨. The technology for production of 1,3-propanediol by the gene engineering has been broken through, especially by the pathway engineering. The cost of production was 25% lower than that by the traditional chemical production. The industrialized production will be realized in the future. Glycerol 3-phosphate dehydrogenase(GPD1), glycerol 3-phosphatase (GPP2), glycerol dehydratase (dhaB) and 1,3-propanediol oxidroreductase (dhaT) are introduced, which construct the super gene engineering for producing the 1,3-propanediol. The key enzyme, dhaB, are analyzed. The effect of the accumulation of shuttle plasmid and glycerol 3-phosphatse on production is introduced. Finally, the focus and the strategy are also discussed for the future.
