试验鸡感染H5N1亚型禽流感病毒后排毒规律的研究

摘  要: 目的 为揭示H5N1亚型禽流感病毒感染试验鸡后体内各组织的排毒规律。方法 将 180只 2 8d非免疫石岐杂鸡随机分为两组 ,每组 90只 ,一组为攻毒组 ,用含H5N1亚型AIV(A/Goose/GD/ 2 / 96 ,对 2 8d非免疫石岐杂鸡LD50 为 10 -5 4× 0 2ml)尿囊液 10 -4× 0 2ml/只经肌肉注射途径攻毒 ,一组接种 0 2ml/只灭菌生理盐水为对照。攻毒后前 4d ,每隔 6h观察试验组和对照组鸡的临床表现 ,每组随机抽取两只剖杀 ,收集脑、肝、脾、肾、胰腺、肌肉、喉头与泄殖腔拭子进行病毒分离 ;攻毒4d后改为每隔 12h取样 ,直至第 11d ,同时采集血清测定禽流感HI抗体效价。结果 试验鸡攻毒 18h后可从泄殖腔及喉头拭子中分离到病毒 ,持续到攻毒后第 7d左右 ;攻毒后 36h可以从脑、肝、脾、肾、胰腺的混合样品中分离到病毒 ,可持续到攻毒后第 10d左右 ; In order to observe the dynamic changes on shedding virus of the experimental infection of chicken with the avian influenza virus (AIV) H5N1 subtype,180 non immunized Siqiza chickens of 28 days old were randomly divided into two groups with 90 chickens in each group,of which one group was challenged with AIV H5N1 subtype(A/Goose/GD/2/96,the LD 50 for non immunized Siquza chickens of 18 days old was 10 -5.4 ×0.2 ml) in a dosage of virus at 10 4×0.2 ml of allantoic fluid intramuscularly,and the other group was challenged with 0.2 ml sterile saline as control.Within the first 4 days after challenging,the clinical symptoms and the pathological changes were recorded every 6 hours,and from which two chickens were taken and killed.The specimens of internal organs such as brain,liver,spleen,kidney,pancreas and muscle as well as the cloacal swab and tracheal swab were taken for virus isolation.and the samples were collected every 12 hours 4 days after challenging till 11days thereafter.At the same time,sera were taken for the determination of anti AIV HAI antibody titres.It was found that AIV H5N1 subtype could be isolated from cloacal and tracheal swabs 18 hours after inoculation and lasted for 7 days,and could also be isolated from the mixed specimens of various viscera 36 hours after inoculation,and lasted for 10 days;no virus could be isolated from the muscle samples
