电穿孔法转移GHRH(1-32)质粒在仔猪肌肉中表达对生产性能的影响

摘  要: 采用pcDNA3.0-GHRH(1-32)质粒,对断奶仔猪肌肉单次注射后电穿孔处理,观察对猪生产性能的影响。采用碱变性法提取pcDNA3.0-GHRH(1-32)质粒并定量。选取32日龄仔猪60头,公母各半,分别注射0(生理盐水)、1.0mg GHRH表达质粒,然后用电穿孔仪电击处理。结果表明,与对照组相比,试验组猪32日龄初始平均体质量无差异(P>0.05),公猪67和98日龄平均体质量分别显著增加26.71%(P<0.05)和21.31%(P<0.05),而母猪呈增加趋势但差异不显著(P>0.05);公猪32~67日龄日增重显著增加47.93%(P<0.05),而母猪呈增加趋势但差异不显著(P>0.05);试验组淘汰率降低20.04%,其中公猪淘汰率降低33.50%;试验组猪80、150和180日龄血清GHRH浓度分别显著增加13.64%(P<0.05)、15.82%(P<0.05)和11.36%(P<0.05),血清中IGF-1浓度分别增加26.70%(P<0.05)、4.31%(P>0.05)和6.04%(P>0.05),且母猪血清浓度增幅高于公猪;血清中其余激素(T3、T4、IL-2、IL-6)和IgG浓度也呈上升趋势等。本试验表明GHRH(1-32)表达质粒可提高猪生产性能。 In the present study,piglets were treated with a single injection of pcDNA3.0-GHRH(1-32)plasmid followed by electroporation to explore its effects on production performance of piglets.The previously constructed plasmid was extracted by using alkaline denaturation method,then plasmid was injected in different dosage(0and 1.0mg)into muscular semime-branosus of piglets(n=60).The results showed that compared with 1.0mg dosage group and the control,the average of piglets body weight were no difference at 32-day-old.The average body weight of the boars were significantly increased by 26.71%(P<0.05)and 21.31%(P<0.05)at 67 and 98-day-old,while the sows were no significant difference(P>0.05).The daily gain of boars were significantly increased by 47.93%(P<0.05)in 32-67day-old,while the sows were no significant difference(P>0.05).The mortality reduced by 20.04%,specially in boars reduced by 33.50%.Piglets serum GHRH concentration were significantly increased by 13.64%(P<0.05),15.82%(P<0.05)and 11.36%(P<0.05),serum IGF-1were increased by 26.70%(P<0.05),4.31%(P>0.05)and 6.04%(P>0.05)in 80,150 and 80-day-old,respectively.Serum(T3,T4,IL-2,IL-6)and IgG concentrations were also increased in test groups.The results above indicate that the injection of GHRH expression plasmid followed by electroporation is a potential way to improve production performance of piglets.
