摘 要: 【目的】检测绵羊X染色体59578440位点在尾型极端差异的阿勒泰羊、湖羊和中国美利奴细毛羊群体中的多态性,并验证该位点是否可作为一个有效的分子标记应用于低脂绵羊新品种培育。【方法】采用PCR-RFLP方法进行SNP分型,并使用卡方检验的方法验证各基因型在群体中的Hardy-Weinberg平衡性。【结果】扩增出了含绵羊X染色体59578440位点SNP的296 bp片段,经AatⅡ核酸内切酶酶切分型后鉴定出了三种基因型(CC、TT和TC)。C/T等位基因的比值显示阿勒泰羊群体中C等位基因属优势等位基因。【结论】X染色体59578440位点SNP可作为阿勒泰羊选育的一个实用性分子标记。 【objective】To detect the polymorphic loci( 59578440) on X chromosome in extremely consonant tail- type Altay,Hu and Chinese Merino populations,and verify whether the SNP can be used as an effective molecular marker in low- lipid sheep breeding. 【Method】PCR- RFLP method was used for SNP genotyping,and Chi- Square test was employed to verify the Hardy- Weinberg equilibrium of genotype groups.【Result】A 296 bp fragment containing the 59579440thloci SNP on X chromosome was successfully amplified,and three genotypes( CC,TT and TC) were identified by AatⅡ endonuclease enzyme cutting. The C / T allele ratio of Altay sheep population indicated that C allele is a dominant allele. 【Conclusion】The SNP located at the 59578440thsequence on X chromosome can be used as a molecular marker in Altay sheep breeding.
