摘 要: 目的:探讨碳源对芽胞杆菌碱性纤维素酶(CMCase)合成的影响,揭示酶的阻遏合成规律。方法:采用分批培养法,改变培养基中基质种类及浓度,定时取样测定CMCase酶活(DNS法)和菌体干重(DCW),以比合成作为产酶水平的指标。结果:碳源种类对酶合成水平影响不大。以葡萄糖为碳源培养,4%浓度时酶合成水平最高,≥5%的高浓度下酶合成才有部分被阻遏。ATP、三羧酸循环中间物对酶合成有一定的阻遏作用。结论:芽胞杆菌EV23产的碱性纤维素酶为组成型酶,酶的抗阻遏合成有一定限度。 Objectives: To investigate the effects of carbon sources on the alkaline cellulase synthesis by Bacillus sp. Methods: Varying the types and concentration of substrates in medium, we measured the enzyme activity (DNS assay) and dry cell weight (DCW) of every sample taken timely from batch cultures. Results: The effects of variety carbon sources on the enzyme productivity was not apparent. Using glucose as carbon source, the enzyme productivity reached the highest at the concentration of 4% . repressed partially when the level reached at least 5%. The presence of ATP and intermidiates in TCA caused the repression of CMCase formation to some extent. Conclusions: The alkaline CMCase by Bacillus sp. EV23 belongs to constitutive enzyme.
